脳は“場所ごと”に老け方がちがった。AIが約1.5万人のMRIで描いた「脳年齢マップ」

脳は、どの場所もほぼ同じスピードで老化していく
AIで場所ごとの脳年齢を測ると、前頭葉・側頭葉は頭頂葉・後頭葉より老けて見えた。

約1.5万人の脳MRIをAIに学ばせ、脳の場所ごとの「年齢」を地図にした研究。前頭葉と側頭葉が、頭頂葉や後頭葉より老けて見えた。
ひとことで言うと脳は全部いっしょに年をとるんじゃなくて、場所ごとに老け方がちがうらしい!
「脳年齢」と聞くと、脳全体にひとつの年齢がついているイメージがありませんか?
でも実は、脳は場所によって老け方がちがうらしいのです。アメリカの南カリフォルニア大学(USC)のチームが、AIに約1万5000人分の脳のMRI画像を学ばせ、脳の小さな区画ごとに「何歳っぽいか」を地図にしました。
すると、おでこの奥の前頭葉と、耳の上あたりの側頭葉が、頭のてっぺん寄りや後ろ側の場所より老けて見えることがわかりました。軽度認知障害やアルツハイマー病の人では、この“老け”がさらに強く出ていました。
01脳に「場所ごとの年齢」をつけてみた
これまでの「脳年齢」研究は、MRI画像から脳全体でひとつの年齢を推定するのが一般的でした。実際の年齢より脳年齢が高ければ「脳が老けている」と考えるわけです。
ただ、ひとつの数字にまとめると、「どこが」老けているのかが見えなくなります。そこで今回のチームは、MRI画像を細かい立方体(ボクセル)の単位で見て、それぞれの場所に年齢をつけるAIを作りました。
02老けて見えたのは「おでこの奥」と「耳の上」
健康な大人の脳では、前頭葉と側頭葉が、頭のてっぺん寄りの頭頂葉や後ろ側の後頭葉よりも、相対的に老けて見えました。
前頭葉は計画を立てたり判断したりする場所、側頭葉は記憶や言葉に関わる場所です。年をとると「段取りが苦手になる」「名前が出てこない」と感じやすいのも、なんとなく納得できますね。
この結果は、昔からの研究ともおおむね合っています。健康な大人の脳の体積を5年間追いかけた2005年の研究では、海馬や、高度な情報処理を担う「連合野」の皮質が大きく縮んだ一方、後頭葉にある一次視覚野はほとんど縮まなかったと報告されています。
さらに、軽度認知障害、アルツハイマー病と進むにつれて、前頭葉と側頭葉の「老け」がどんどん強くなるパターンも見られました。アルツハイマー病で早くから変化が出るとされる場所は、軽度認知障害の段階でも、健康な人より有意に年上に見えたといいます。
そして、場所ごとの「標準からのズレ」は、その場所が担う認知機能のテスト成績とも関係していました。AIの地図が、ただの絵ではなく、頭の働きと結びついているというわけです。

03補足:大学の発表では「左右の差」にもふれていた
論文の要旨には書かれていませんが、南カリフォルニア大学の発表(USC Today)によると、健康な大人では右半球が左半球よりわずかに老化が進んで見える傾向があり、それは右利きでも左利きでも同じだったそうです。
ただし、この左右差がどのくらいの大きさなのか、どんな条件で確かめたのかは、要旨にも大学の発表にも数字が出ていません(この記事では、論文本文の数字までは確認できていません)。「右脳から先に老ける」と言い切れる話ではない、と考えておくのが安全です。
04どんでん返し:「右脳から老ける」説は、実は昔からの論争
じつは「右半球のほうが早く衰える」という考え(右半球老化説)は、昔から心理学で議論されてきました。
ところが、2002年にデューク大学のチームが証拠をまとめた総説では、右半球老化説を支持する結果はまちまち。むしろ、高齢になると前頭葉の活動の左右の偏りが小さくなる(HAROLDモデル)という説明のほうが、脳画像の証拠とよく合うとされました。ただしこの総説は、2つの説は両立しうる(前頭前野と、それ以外の場所で別々に当てはまるかもしれない)とも書いています。
さらに2021年、ノルウェーなどのチームが複数の追跡調査グループの大人を何年も追いかけた研究では、20歳ごろに左右差がはっきりしていた高次の領域で、もともと厚かった側の大脳皮質が速く薄くなり、左右差が失われていくことが報告されています。加齢で起きているのは「右が負ける」ことより、「左右の個性がならされていく」ことのほうなのかもしれません。
大学の発表にある「右がわずかに年上」という話が、こうした流れとどうつながるのか。答えはまだ出ていません。
05まとめ
脳は「まるごと一緒に」年をとるわけではなく、前頭葉と側頭葉が、頭頂葉や後頭葉より老けて見える。AIが描いた脳年齢マップは、そんな凸凹を見せてくれました。
将来は、全体の脳年齢だけでなく「どこが老けているか」から、認知症のサインを早く見つけられるかもしれません。あなたの脳のどこが一番“若い”のか、ちょっと気になりませんか?

Ask how old a brain is, and most studies will give you one number. Researchers at the University of Southern California tried something more detailed. They trained a deep-learning model on MRI scans from 14,748 cognitively normal adults, then asked it to estimate the age of every small cube of brain tissue. The resulting maps were uneven.
In healthy people, the frontal and temporal lobes, which support planning, memory and language, appeared older than the parietal and occipital lobes. When the model was applied to 354 people with mild cognitive impairment and 529 with Alzheimer's disease, this frontotemporal pattern grew stronger with each stage. Regions that drifted further from the normal pattern were linked to weaker scores on related thinking tests.
The university added that the right hemisphere looked slightly older than the left, regardless of handedness. That fits an old idea called the right hemi-aging hypothesis, but a 2002 review found the evidence for it mixed. Most of the scans came from a single point in time, so the maps mainly compare different people and cannot show that anyone's right hemisphere is aging faster. According to the university, the next step is to follow the same people for years.
